Graham, 2 wires for each speaker. I think Blue/Pink and Blue/Black for the left, and Slate/Pink and Slate/Black for the right. Normal wiring is direct from rear of radio to speakers. How does your head unit interface to the wiring? I doubt the Rover plug will be a direct fit so are the wires spliced or is there an adaptor in use? Could there be a disconnection at the adaptor?
Checking the wiring end to end isn't a big problem as long as you have access to a multimeter. Disconnect the wiring at the speakers then either connect the car +12 volt to each wire at the radio end and check from wire to earth at the speaker end, or, put a small battery across the pair of wires at the radio and check for battery voltage between the wires at the speaker end.

ok, well if you've established the head unit is good and the speakers are good then it has to be the wiring. Fuses dont come into it as if the radio lights up and appears to work then the fuses are fine.
Take the dashboard out. Then on the passenger side, where the passenger door check assembly retracts into the body you'll see the wiring for the rear of the car disapeaing up into the A pilar. If they chaff though anywhere they do it here. They can get caught in the door checker or just rub through on the hole in the body.
You can either splice the wires back together or you can cut them off at the back of the ISO plug on the stereo and run new ones under the carpet to the boot.
However i would check for chaffing where i describe as this is also the route for your screenwash wiring as well as the rear lighting and fuel pump, so if the speaker wires have broken then there is a good chance something else is likely to.

hb, how do you remove the wooden dash on an MPi mini? This is the first mini I've owned with this luxury!

Easy,
- undo the bolt that secures the steering column to the lower dash and let the column droop a bit.
- Remove the radio
- remove the airvents (just unscrew the outer rings and the twist out the vents)
- 10mm spanner and undo the nut you can see at the bottom of the radio hole.
- then either end of the dash under the bottom dash rail there is another 10mm nut at each end. (small ratchet with a 3" extension is best for these)
- Open both doors
- Pull the dash out towards you while unhooking the bottom brackets which you took the nuts off.
- Unplug speedo cable and electrical cables.
While you have it out, remove the plastic box for the glove compartment and glue a lining inside, i used felt from a local craft shop.
Also cover the back of the glove box with self adhesive flashing tape (roofing section in B&Q)
remove the top serated bracket and cut off the serated bit, then fix it to a small metal angle bracket (again 50p in B&Q)
This will stop the dash jumping up and down and anything you put in the glovebox rattling like hell.

Right just to update this thread.....
Took the time to take the dash out over the Easter break which was suprisingly easy (cheers hb). I checked the wires (colours as you described Dave) but they seemed to be okay. At this point (I've no hair left to pull out!) I bit the bullet and called an auto stereo type bloke. £20 later (Grrr) he confirmed that my basic understanding of electrics was correct, the speakers are okay, the wires to the speakers are okay and there is an output from the stereo! But after checking with another stereo there has been some sort of internal problem in the head unit (he couldn't explain why there was an output but no sound at the speakers)!
So, now at least I have an answer! Can anyone recommend a head unit for around £100? Cd player and preferably iPod connection too.

(Oh, and took your advise hb and trimmed the glovebox at the same time. Suffice to say I was bloody pleased with the result, I'll try and post a pic I'm so proud!)
